Uppsala university has excelled in ribosome research over decades and already has a strong environment in the area of bacterial protein synthesis. We aim here at creating a novel type of ribosome research, in which biochemistry, structural biology, molecular computations, molecular biology, single molecule spectroscopy in vitro and in vivo along with systems biology modeling of living cells are fully integrated from experimental design to data analysis. One part of the proposal is to develop or adapt powerful techniques for universal studies of ribosome function - high resolution and time resolved cryo-EM, computational methods for structure based free energy calculation, biochemical method for analysis of complex multistep processes and single molecule-single cell tracking with microscopy.  With those tools we will expand the present studies of the accuracy and efficiency of codon reading by transfer RNAs and release factors in bacteria to eukaryotes and organelles. This scientific intent is particularly challenging since the biochemistry of protein synthesis in other than bacterial systems is poorly developed, but we have preliminary results that promise to open these systems for quantitative ribosome research. We will also use this interdisciplinary environment to train a next generation of researchers through laboratory rotation programs, courses and seminar series in which internationally leading scientists will participate along with the five PIs of the proposal. .
